Human Cytomegalovirus-Encoded microRNAs Can Be Found in Saliva Samples from Renal Transplant Recipients
The human cytomegalovirus infection (HCMV) is general after kidney transplants and may have a long-term effect. HCMV can be measured directly by DNA virus or indirectly through the host of the immune response. Microrna encoded HCMV (MIRNA) can change the pathobiology of HCMV infection and contribute to the development of HCMV. MIRNAS encoded HCMV can be detected in the blood but has not been sought with saliva. We investigate the samples of saliva from 32 recipients of kidney transplant (RTR) and 12 seropositive health controls for whom immunological data is available.
Five Mirnas encoded HCMV (MIR-UL112-5P, MIR-US5-2-3P, MIR-UL36, MIR-US25-2-3P and MIR-UL22A) are searched using the primary probe test. HCMV Mirna species detected in saliva from 15 RTR and 3 healthy controls, with the most commonly detected MIR-US5-2-3P. The presence of HCMV MInnas is related to an increase in the T-cell response to HCMV IE-1 on RTR, shows a frequent reactive link from HCMV.
Cytomegalovirus endotheliitis after penetrating keratoplasty
Cytomegalovirus (CMV) – The chain corneal endothelial is the inflammation of the corneal endothelia caused by CMV. This can happen de novo or after ocular surgery in healthy individuals. In patients who have undergone Keratoplasty, the diagnosis of appealing viral endotheliitis and the rejection of the graft regarding the immune system challenges because of similar clinical findings.
Here we report a patient who underwent keratoplasty penetrating and using local and systemic immunosuppressive agents because of the history of the previous graft rejection. In the 4th postoperative, eye examination revealed local corneal edema, coin-shaped kereal deposits, and increased intraocular pressure, consistent with viral endotheliitis. The polymerase chain reaction reveals CMV-DNA amplification in aqueous humor sample. Valganciclovir treatment began and the symptoms improved in 2 months. It must be remembered that local or systemic immunosuppressants are used after keratoplasty can trigger CMV reactivation. Anti-CMV treatment must begin immediately in patients with coin-shaped keratic deposits.
Cytomegalovirus (CMV) management in alogenic hematopoietic cell transplants: Predictor pre-transplant survival, reactivation and spontaneous permits
Cytomegalovirus (CMV) reactivation is a complication that often occurs after alogenic hematopoietic cell transplants (ALLOHCT). We analyzed 159 ALLOHCT receivers with 4409 quantitative viral viral to determine the pre-transplanted predictor of CMV reactivation, clinically significant CMV infection (CS-CMVI, defined as Viral Load CMV> 1000 IU / ml), CMV disease, spontaneous cmv cleaning kinetics and survival Using a standard pre-emptive therapy approach to identify risk groups to target prevention strategies.
The most common CS-CMVI in non-related donor donor transplants (URD). Spontaneous CMV permits occurred in 26% of patients who reached Viral Load 56-137 IU / ML, 6% in 138-250 IU / ML and in 1 patient> 250 IU / ml. The median time between the first CMV reactivation (> 56 IU / ML) and Viral Load> 250 IU / ML is 13 days, while the time from the first viral load> 250 IU / ML to reach the Loading Vial> 1000 IU / ML 4 days. CS-CMVI is associated with a significant increase in non-relapse mortality (NRM) in multivariate analysis. Overall, this study shows that URD D / R + recipients are at high risk for mortality related CSVI and CMV, and are potential candidates for targeted CMV prophylaxis. CMV spontaneous cleaning outside the 250 IU / ML viral load is rare, indicating that this can be used as the appropriate threshold for starting pre-emptive therapy.

Cytomegalovirus-specific-specific-t-cell-specific-specific-cell receptor receptor after the alogenic stem cell transplantation
Cytomegalovirus (CMV) infection is the main complication during the alogenic stem cell transplantation (allo-sct). However, the mechanism of adaptive immunity which encourages this remains unclear. To define the initial immunological response against CMV after transplantation, we use the next generation sequencing to check the CD8 + / CMV Tetramer + cell (CMV-CCV) cell receptor (CMV-CTLS) in a peripheral blood sample obtained from 16 Allo-SCT receivers with HLA , -A * 24: 02 at the time of CMV reactivation.
In most patients, TCR Beta CMV-CTLS repertoar is very tilted (Median Inverse Simpson’s Index: 1,595) and, 15 of the 16 patients sharing at least one TCR-beta clone with ≥ 2 patients. TCR shared dominant in 12 patients and, two clonotypes were divided around half of the patients. The similarity analysis shows that CDR3 sequences from TCR together are more similar than TCR which is not divided. TCR beta repertoir from CMV-CTL in 12 patients was also analyzed after 2-4 weeks to characterize the short-term dynamics of the TCR repertoire. In ten patients, we observe the persistence of applicable clones. In two other patients, the TCR repertoire became more diverse, the main clone declined, and new private clones later emerged. These results provide substantive instructions to understand immunological behavior towards CMV reactivation after allo-sct.
Protein kinase autophagy-initiate ulk1 phosphorylation human cytomegalovirus tegument protein pp28 and regulate the release of an efficient virus
Autophagy is a catabolic process that contributes to intrinsic cellular defense by degrading viral or protein particles, but some viruses hijack this path for their own benefit. The role of autophagy during the replication of human cytomegalovirus (HCMV) has not been clarified earlier. Utilizing Sirna-based filtering, we observed that the depletion of many autophagy related proteins resulted in reduced virus releases showing the requirements of autophagy related factors for efficient HCMV replication.
In addition, we can show that the autophagy-initiation of Serine / Threonine-Protein Kinase ULK1 and other constituents of the UK1 complex have been regulated during the initial infection and remain regulated throughout the replication cycle. We show that indirect disturbances with ULK1 through the inhibition of the release of the UPSTRAM regulator regulator Virus Regulator. Furthermore, this result is verified by the direct revocation of Ulk1 Kinase activity that uses the Special Kinase Inhibitor Ulk1 SBI-0206965 and ULK-101.
Analysis of viral protein expression with the existence of an ULK-101 revealed the connection between the cellular kinase ULK1 and Viral Tegument Protein PP28 (Pul999) and we identified PP28 as a novel virus substrate with a vitro kinase test. In the absence of ulk1 kinase activity, a large PP28- and PP65-positive structure can be detected in the cytoplasm at the end point of the infection.

Description: Proteins encoded by the v-Rel viral oncogene and its cellular homolog, c-Rel,are members of a family of transcription factors that include the two subunits of the transcription factor NFκB (p50 and p65) and the Drosophila maternal morphogen, dorsal. Both proteins specifically bind to DNA sequences that are the same or slight variations of the 10 bp κB sequence in the immunoglobulin κ light chain enhancer. This same sequence is also present in a number of other cellular and viral enhancers. The DNA binding activity of NFκB is activated and NFκB is subsequently transported from the cytoplasm to the nucleus in cells exposed to mitogens or growth factors. cDNAs encoding precursors for two distinct proteins of the same size have been described, designated p105 and p100. The p105 precursor contains p50 at its N-terminus and a C-terminal region that when expressed as a separate molecule, designated pdI, binds to p50 and regulates its activity. |
